A study in rats showed that the clearance of both Sildenafil (Viagra) and Sildenafil-HCl in the blood plasma is significantly different in rats with and without chronic renal disease. Sildenafil had little effect on the plasma levels of Sildenafil-HCl in both chronic renal disease and control groups, while Viagra had a significant effect on the plasma levels of both drugs in both chronic renal disease and control groups. Therefore, we suggest that Sildenafil and Viagra may be responsible for the effects of Sildenafil in the rat, which is consistent with the results of the present study.
Concomitant administration of Sildenafil and Sildenafil-HCl reduced the plasma levels of both drugs in rats, with no significant difference between chronic renal disease and control groups. This may be due to the fact that both drugs have different pharmacokinetic profiles, with the most active Sildenafil being more active in the renal tubules and the least active Sildenafil in the lumen. This may explain the differences in the pharmacokinetic profile between the drugs, which are not fully elucidated by the present study.
